Casma Overview

Casma Therapeutics is harnessing autophagy by developing a novel degradation technology to open new target areas for drug discovery and development that will profoundly impact the lives of patients. Autophagy is a conserved cellular process that contributes to overall organismal health, but when autophagy is perturbed, inefficient autophagic flux contributes to numerous diseases. By selectively boosting autophagy, Casma expects to be able to arrest or reverse the progression of several diseases such as neurodegeneration, metabolic disorders, inflammation, and muscle degeneration. Casma was launched in 2018 by Third Rock Ventures and recently completed a Series B financing led by The Column Group with participation by new investors Eventide and Schroeder Adveq.

The Role

We are seeking a highly motivated Associate Scientist/Senior Associate Scientist to join a cross-functional group focusing on target ID/validation, in vitro efficacy model generation, and assay development/small molecule screening in disease-relevant cell systems. This team member will  deploy cutting edge gene-editing technologies in a variety of iPSC-derived cell types (e.g., neurons and myoblasts) to support both exploratory biology and drug discovery efforts. Responsibilities

  • Establish, maintain and differentiate iPSCs and primary cells to desired cell types.
  • Perform CRISPR-based gene-editing and construction of stable cell lines in iPSC lines to drive target ID/validation and assay development across multiple therapeutic programs.
  • Develop and optimize quantitative in vitro efficacy assays in iPSCs and/or primary cells using a variety of techniques (genetic engineering, high content imaging, FACS, HTRF, luminescence, etc).
  • Actively participate on a dynamic and collaborative team focused on developing new medicines for patients in need.
  • Independently interpret data and recommend next steps.
  • Contribute to a fun, intellectually stimulating and fast-paced work environment.

Requirements + Qualifications

  • Bachelor’s or Master’s degree in biology or related discipline plus 5-7 years’ experience conducting high quality science in an industry setting.
  • Extensive experience with cell culture/differentiation, CRISPR based genome engineering, stable cell line generation, and disease modelling in iPSCs.
  • Genetic screens and/or in vitro assay development experience (medium-to-high throughput) in iPSC systems is preferred.
  • Proficient with flow cytometry, western blot analysis, and high-content imaging assays.
  • Significant knowledge/experience with CNS aggregate models is a plus.
  • Demonstrated excellence in independently planning, executing, and analyzing experiments.
  • Excellent organizational skills and attention to detail.
  • Strong interpersonal collaboration and communication skills.
