Job Description

Casma Therapeutics is harnessing the natural cellular process of autophagy to open vast new target areas for drug discovery and development that will profoundly impact the lives of patients. Autophagy is a conserved cellular process that contributes to overall organismal health. Inefficient autophagy flux or defects in lysosomal homeostasis are observed in numerous pathologies, e.g., neurodegeneration, metabolic disorders, inflammation, and muscle degeneration. Casma uses several approaches to intervene at strategic points throughout the autophagy-lysosome system to improve the cellular process of clearing out unwanted proteins, organelles and invading pathogens. By boosting autophagy, Casma expects to be able to arrest or reverse the progression of lysosomal storage disorders, muscle disorders, inflammatory disorders and neurodegeneration, among other indications. Casma was launched in 2018 by Third Rock Ventures and is based in Cambridge, Mass.

The Role

We are seeking a talented scientist who is eager to contribute to the build of Casma’s autophagy degrader platform. The candidate is expected to be a team player who will contribute to both our drug discovery and discovery biology engines to inform molecular development and pathway biology. This future Casmaniac will have experience in drug discovery and the desire to develop cell-based high throughput assays to drive key drug discovery workflows, progress hit validation and lead optimization, and show in vitro efficacy. While experience in autophagy is not necessary, cell biology experience in an industry setting is critical.  Flexibility and learning agility are a must, as we are looking for someone to develop and execute cell-based assays across diverse targets and in vitro disease models.  We are seeking a qualified candidate who will share our excitement about the science, contribute to our culture, and honor our commitment to patients.

Responsibilities

  • Create impact for lead Casma programs by developing plate-based assays using high content cellular imaging platforms, flow cytometry and multimode plate-based reader systems
  • Utilize instrumentation and robotics to aid in high throughput (384-well) methodologies
  • Develop and characterize appropriate cell lines and reporters necessary for assay development
  • Where necessary, work with primary or patient derived cells to assess target engagement and to develop in vitro disease efficacy models; interface with PD marker and in vivo teams
  • Develop an understanding of the autophagy pathway to independently drive experiments
  • Independently interpret data, recommend next steps, and engage colleagues for advice/questions
  • Actively participate on a dynamic and collaborative team focused on patient outcomes
  • Demonstrate the ability to take the lead and prioritize sensitive projects
  • Ability to write detailed protocols and effectively troubleshoot issues in order to partner with CROs
  • Other duties, as assigned.

Requirements + Qualifications

  • Bachelor’s degree in biology or related discipline plus 5 years of experience or Master’s degree plus 3-5 years of experience conducting high quality science in an industry setting
  • Proficient in mammalian cell culture techniques such as microscopy, antibiotic selection, clonal cell isolation, transfection, infection, virus preparation, and cell line generation
  • Experience with molecular biology techniques such as cloning methods, qPCR, gel electrophoresis, bacterial culture
  • Biochemical experience such as western blotting, immuno-precipitation, immunocytochemistry, and sample preparation
  • Experience with plate-based assay (e.g. Luciferase, AlphaLISA, HTRF, MSD, ELISA, etc)
  • Excellent organizational skills and attention to detail
  • Clear and effective verbal and written communication skills
  • Ability to successfully participate in a highly creative and enthusiastic start-up environment
  • Exemplify a patient-first focus, courage, integrity, drive, and respect for all team members

Additional preferred skills

  • Expertise in high content imaging and analysis is highly preferred
  • Prior use of lab automation and liquid handling systems is a plus
  • Knowledge of primary cell culture and/or development of in vitro disease models is also a plus
